P-11 Response of pancreatic ductal adenocarcinoma to gemcitabine using a human organotypic slice culture model

The organotypic tumor slice culture model resembles the in vivo situation of tumors. It may serve as a good ex platform for response to anti-tumor therapies. We aimed evaluate pancreatic ductal adenocarcinoma (PDA) cells and helper, cytotoxic, regulatory T gemcitabine using human PDA model. tissues were obtained from patients who operated on resection. A vibratome sliced tumors, slices cultured until 5 days with 0.1 μM, 1 10 μM or 2 days. Tumor responses evaluated by number proportion Ki-67 caspase-3 positively expressed tumor. CD4+, CD8+, FOXP3+ analyzed immunohistochemistry staining. Gemcitabine treatment exerted proportional cytotoxic effects terms cell number, cleaved expression.
